The insulin receptor (INSR) is a heterodimeric protein complex that has an intracellular subunit, which is disulfide-linked to a transmembrane segment. The insulin ligand binds to the INSR and initiates molecular signaling pathways that promote glucose uptake in cells and glycogen synthesis. Insulin binding to INSR induces phosphorylation of intra-cellular tyrosine kinase domains and recruitment of multiple SH2 and SH3 domain-containing intracellular proteins that serve as signaling intermediates for pleiotropic effects of insulin. B-cell lymphoma/leukemia 10 (BCL10) is a signaling protein that promotes apoptosis, pro-caspase-9 maturation, and activation of NF-kappa-B via NIK and IKK. BCL10 contains an N-terminal caspase recruitment domain (CARD), which is found in a number of apoptotic regulatory molecules. BCL10 is involved in the adaptive immune response, and may be an adapter protein between upstream TNFR1-TRADD-RIP complex and the downstream NIK-IKK-IKAP complex. A BCL10 translocation is recurrent in low-grade mucosa-associated lymphoid tissue (MALT lymphoma). Mammaglobin is a 93 amino acid glycoprotein with homology to other secretoglobin-uteroglobin family members. It was originally identified as a breast cancer restricted biomarker by differential screening. Mammaglobin related to secretoglobin family that includes human uteroglobin and lipophilin. Mammaglobin antibody stains cytoplasm of normal breast epithelial cells as well as primary and metastatic breast carcinomas. Mammaglobin expression is absent in prostate, kidney, colon, rectum, small intestine, stomach, pancreas, lung, and thyroid tissues. Cadherin 17, also known as LI cadherin (for liver-intestine cadherin), is a member of the cadherin family of calcium-dependent adhesion molecules. Cadherins mediate cell-cell interactions via hemophilic binding, and are critical to the maintenance of tissue structure and morphogenesis. Cadherins each contain a large extracellular domain at the amino terminus, which is characterized by a series of five homologous repeats, the most distal of which is thought to be responsible for binding specificity. The relatively short carboxy terminal, intracellular domain interacts with a variety of cytoplasmic proteins, including beta-catenin, to regulate cadherin function. Cadherin 17 expression is restricted to liver and intestine tissues and is specifically localized to the basolateral domain of hepatocytes and enterocytes. Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI 6.0) subfamilies. The v-Myc oncogene, initially identified in the MC29 avian retrovirus, causes myelocytomas, carcinomas, sarcomas and lymphomas, and belongs to a family of oncogenes conserved throughout evolution. In humans, the family consists of five genes: c-Myc, N-Myc, R-Myc, L-Myc and B-Myc. Amplification of the N-Myc gene has been found in human neuroblastomas and cell lines. Its amplification correlates well with the stage of neuroblastoma disease. Immunological studies have shown that the human N-Myc gene encodes a nuclear phosphoprotein that exhibits relatively short (30 min) half life in vivo. Spectrin is an actin crosslinking and molecular scaffold protein that links the plasma membrane to the actin cytoskeleton, and functions in the determination of cell shape, arrangement of transmembrane proteins, and organization of organelles. Spectrins function as membrane organizers and stabilizers, composed of non-homologous α andβ chains, which aggregate side-to-side in an anti-parallel fashion to form dimers, tetramers and higher polymers. Spectrin α I and spectrinβ I are present in erythrocytes, whereas spectrin α II (also designated fodrin α) and spectrinβ II (also designated fodrinβ) are present in other somatic cells. This antibody recognizes a protein of 39 kDa, which is identified as Aurora kinase B (or Aurora B). Aurora B is a serine/threonine protein kinase and component of the chromosomal passenger complex (CPC), a complex that acts as a key regulator of mitosis. It is important for accurate chromosome segregation, cytokinesis, protein localization to the centromere and kinetochore, correct microtubule-kinetochore attachment, and regulation of the mitotic checkpoint. Aurora B forms a tight complex with inner centrosome protein and survivin. Inactivation of any of these proteins causes similar defects in chromosome segregation.
